Tryptophane Metabolism I. the Production of Kynurenic Acid from Tryptophane Derivatives* by Clarence P. Berg

چکیده

Recent work (Berg, Rose, and Marvel, 1929-30) has shown that the essential amino acid, tryptophane, may be rendered unfit for supporting growth, either by benzoylation or by condensation with formaldehyde. The formaldehyde condensation product has been employed also by Jackson (1929) with a similar result. On the other hand, neither esterification with ethyl alcohol nor acetylation caused any detectable decrease in its growth-promoting ability. Although the ethyl ester hydrochloride used was shown to be very readily split by enzyme extracts, the acetyl derivative appeared to undergo cleavage only very slowly and incompletely. Inasmuch as tryptophane undergoes a series of react, ions in certain animal organisms by means of which it is converted into the quinoline derivative, kynurenic acid (Ellinger, 1904), it seemed of interest to employ the output of the latter product in the urine, following the administration of the tryptophane derivatives mentioned above, as a means of obtaining evidence regarding their cleavage in vivo. If the derivative in question should undergo enzymolysis either in the alimentary tract or in the body proper, the tryptophane liberated would cause an increase in urinary kynurenic acid elimination. For the purpose of demonstrating the production of kynurenic acid after tryptophane administration, the dog and the rabbit are

The Influence of Optical Isomerism on the Utilization of Tryptophane, Histidine, and Lysine for Growth in the Mouse*

Interesting differences have been noted in the influence of optical isomerism on the utilization of the various essential amino acids by the rat for growth. Thus, when d(+)-tryptophane is added to a diet otherwise deficient in tryptophane it promotes growth which is quite as rapid as that induced when I( -)-tryptophane is supplied (3, 21); under analogous conditions d(+)-histidine is somewhat l...

The Availability for Growth of N-methyl- Tryptophane Administered as Its Acetyl Derivative

Investigation of the metabolic fate of acylated amino acids has disclosed many instances of interesting selective action by the animal organism. Magnus-Levy (1) found it possible to recover t,he unaltered benzoyl derivatives of several amino acids, often in excellent yield, from the urine of animals injected with these compounds. On the other hand, formyl derivatives, as represented by formylgl...

The effect of tryptophane on the excretion of nicotinic acid derivatives in humans.

It has been shown by Krehl et aZ.l that tryptophane may replace nicotinic acid as a growth factor for rats receiving a corn diet. Rosen et al2 have demonstrated that the administration of tryptophane to rats increases the excretion of nicotinic acid derivatives. Experiments reported here with human subjects show that the administration of tryptophane leads to an increased excretion of N1-methyl...

The effect of tryptophane on the urinary excretion of nicotinic acid in rats.

Krehl et al. (1,2) have observed a growth retardation in rats when large amounts of corn grits were included in a low protein ration. The addition of either Z(-)-tryptophane or nicotinic acid corrected the deficiency and allowed normal growth to proceed. The beneficial action of tryptophane has also been observed in nicotinic acid deficiency in the chick (3) and the mouse (4). Although recent r...

The abnormality of tryptophane metabolism in children with mongolism.

There is evidence (Gershoff, Hegsted and Trulson, 1958; Jer6me, Lejeune and Turpin, 1960; O'Brien, Groshek and Streamer, 1960) that children with mongolism have a defect in tryptophane metabolism which is characterized primarily by a diminished urinary excretion of xanthurenic acid. The purpose of this study was to confirm this observation using more exact techniques and to attempt to specify t...
